About the project

About the Project RAC1 is a small GTPase protein that functions as a key signaling node downstream of various microenvironmental signaling pathways. RAC1B, a RAC1 variant with an extra19 amino acid insert, is overexpressed in various cancers, including breast, colorectal and lung cancers. This project will seek to unravel the differences in signalling mechanism of RAC1 and RAC1B leveraging structural biology, high-throughput 19F NMR, advanced molecular simulation, combined with the design, synthesis and biophysical evaluation of small chemical tool compounds that bind tightly and selectively to RAC1B over RAC1. Thus, this interdisciplinary project combines computational, chemical and biophysical aspects to offer an exciting research opportunity at the experimental medicine and enzymology interface. This project aligns with UKRI skills priorities by fostering interdisciplinary expertise at the intersection of data science, human health, and biology. The student will gain proficiency in structural biology, high-performance computing, and molecular modeling, applying advanced algorithms to analyse complex biological data. These computational and quantitative skills are essential for modern health research and align with the MRC’s emphasis on developing quantitative skills and use data-driven approaches to understanding disease. The student will also develop expertise in analytical biophysics, organic synthesis, and cell biology, ensuring a multidisciplinary skill set that bridges computational and experimental sciences. This discipline-spanning approach not only enhances UK research competitiveness but also equips the student with versatile skills for diverse career paths. Eligibility : The standard academic entry requirement for this PhD is an upper second-class (2:1) honours degree (or international equivalent) in Chemistry or Biochemistry OR any upper-second class (2:1) honours degree and a Master’s degree at merit (or international equivalent) in Chemistry or Biochemistry. Previous research experience in structural biology, organic synthesis, or computational chemistry is desirable. This project will remain open until filled.
